Casing material, the substrate in the upper layer used in mushroom cultivation (Agaricus bisporus) is always infected with bacterial - and can also be infected with fungal pathogens. Disease incidence is dependent on various factors including moisture and temperature, but there is also a microbiological component. To better understand the key factors determining disease outbreaks, detailed studies will conducted on biotic and abiotic factors of casing material during mushroom cultivation.
The aim of the project is to identify indicators determining resistance in casing material against important diseases in mushroom cultivation (Mycogone perniciosa (wet bubble disease) and Pseudomonas sp. (bacterial blotch)). The project comprises the following elements:
- Detection methods: development and evaluation of TaqMan assays for quantitative detection of blotch causing Pseudomonas species and Mycogone perniciosa
- Investigation on the distribution of micro-organisms in casing material and the basic materials using microbiome analysis (MiSeq amplicon sequencing) and dilution plating. Isolates will be identified (16S rDNA analysis) and tested for antagonism against the pathogens
- Optimisation of the bioassay by supplementing increasing densities of the pathogen to the casing material. During mushroom growth the population dynamics of the pathogens will be studied with TaqMan assays.
- Evaluation of casing materials of different compositions for their resistance against the pathogens. Physico-chemical properties and microbiome studies will be conducted to find indicators for disease suppression.
- Stability of the suppression. Casing materials with a high and low suppression will be mixed in different ratios to study disease suppression of mixtures.
- Varies mushroom cultivars will be tested for resistance against Mycogone and Pseudomonas
