Needles in a haystack: Pursuing directed evolution of CRISPR-Cas protein for improved genome editing
Summary
CRISPR-Cas proteins have revolutionized genome editing. Nevertheless, optimization and tailoring of CRISPR-Cas systems are still necessary for optimal genome editing. In this thesis we pursued to develop new methods for the optimization of CRISPR-Cas proteins and applied already established methods aiming to improve a well-described Cas9 system. The described research finds significance in methods development and protein optimization and characterization.
