In this project, we want to acquire more fundamental knowledge on CARF/SAVED proteins (ancillary effector proteins of type III CRISPR-Cas systems) and potential applications in biotechnology. To gain more insight, we will carry out in silico analyses to make a short-list of interesting CARF/SAVED protein candidates, produce them in E. coli and perform in vitro experiments. Furthermore, in vivo studies will be performed, by genetic engineering techniques, to mimic their native function and re-purposing into useful application tools. Ultimately, we want to produce a (non-)living cellular biosensor, using type III CRISPR-Cas systems as a RNA-sensing device and CARF/SAVED proteins for signal output.
